Tissue Culture Acclimation: The First 21 Days

Tissue culture plantlets arrive straight from a sterile jar. For 21 days they need a gentle path from 100 percent humidity to normal room air. This is the schedule we give every new seller.

This is a general guide. Every space is different. Light, temperature, humidity, and your mix all change how plants respond. Watch your plants and adjust the schedule to what they show you.

Before the box arrives

  • Set up a clear tub with a lid, or a humidity dome over a tray. One tub per variety keeps things simple.
  • Use a light, airy mix: sphagnum moss, perlite, or a fine bark mix. No garden soil.
  • Place the tub under bright, indirect light or a grow light. No direct sun at any point in the 21 days.
  • Keep the room warm and away from cold drafts and heater vents.

Day 1: unbox and pot

  • Record an unboxing video before you open the box. A claim needs it.
  • Rinse any gel off the roots with room-temperature water.
  • Pot each plantlet so the roots are covered and the leaves are above the mix. Water lightly.
  • Close the lid fully. Humidity inside should be near 100 percent.
  • Photograph any plant that arrived damaged and send the photo with your video the same day.

Days 2 to 7: closed lid

  • Keep the lid closed. Open it once a day for one minute to let in fresh air.
  • Keep the mix damp, never wet. Mist the leaves if they look dry.
  • A leaf that melts or yellows this week is usually a humidity drop. Close the lid and check the light.

Days 8 to 14: crack the lid

  • Prop the lid open about one centimeter. Each day, open it a little more.
  • Watch for new leaves. A new leaf means the plant has started making roots for open air.
  • If leaves droop, close the lid for two days and start again.

Days 15 to 21: lid off

  • Remove the lid for a few hours a day, then for the full day, then overnight.
  • By day 21 the plant should hold its leaves in normal room humidity.
  • Now it is a plant you can pot up, price and sell.

Common mistakes

  • Direct sun. It cooks a plantlet in an hour.
  • Wet mix. Roots that sit in water rot. Damp is the target.
  • Opening the lid too fast. If in doubt, go slower.
  • Skipping the unboxing video. Without it there is no claim.
